The KeyTec® TR‑FRET Human IL‑17AF/IL‑17RA Binding Assay Kit is designed for screening small‑molecule inhibitors, peptides, or antibodies that block the interaction between IL‑17AF and IL‑17RA proteins. This kit is based on KeyTec® TR‑FRET technology, offering advantages such as simple and rapid operation and high accuracy.
As illustrated in the schematic diagram below, KeyTec® TR‑FRET Solar Eu1‑conjugated Streptavidin (abbreviated as SA) is pre‑complexed with biotinylated homodimeric Human IL‑17AF protein (AVI‑Biotin). The IL‑17RA protein is conjugated with KeyTec® TR‑FRET LA2 acceptor. When IL‑17AF binds to IL‑17RA, Solar Eu and LA are brought into close proximity, enabling energy resonance transfer between the donor and acceptor upon external light excitation. The degree of interaction between IL‑17AF and IL‑17RA is determined by measuring the signal intensity at 665 nm. Compounds, peptides, or antibodies that block the IL‑17AF/IL‑17RA interaction will result in a decrease in the TR‑FRET signal.
